Most staphylococci are now resistant to benzylpenicillin because they produce a penicillinase. The penicillinase resistant penicillins are, as their name suggests, resistant to the action of this enzyme and are therefore indicated in infections caused by penicillin resistant staphylococcus.

The main antibacterial agent in this class is Flucoxacillin.
