Lofepramine is a tetracyclic which is thought to be less cardiotoxic and to have fewer anticholinergic side-effects than the older tricyclics like imipramine.

The dose ranges from 70-210 mg per day.

It is degraded to desmethylimipramine, an active metabolite of imipramine, and so shares some of its properties.
