Citrate synthase catalyzes a non-equilibrium, flux-generating reaction.
It is stimulated by oxaloacetate. This is important during exercise because the enzyme pyruvate carboxylase produces significantly more oxaloacetate.
It is inhibited by citrate. Exercise produces a greater proportionate activation of the enzymes isocitrate dehydrogenase and oxoglutarate dehydrogenase. Hence, there is a relative reduction in the amount of citrate and a relative disinhibition of citrate synthase.
